Analysis of samples by flow cytometry

The service is provided for quantitative and/or qualitative analysis of cell or particle samples by flow cytometry.

 

Implementing

Flow cytometry is a biophysical technology based on the use of laser light, used in the counting and classification of cells or particles according to their morphological characteristics, presence of biomarkers, and in protein engineering. In the flow cytometers, cells suspended in a fluid pass through a very thin transparent tube on which a thin beam of laser light hits, the light transmitted and dispersed by the passage of cells through the tube is collected by means of detection devices, allowing inferences as to the size and complexity of the cells, and the detection of fluorescence. It also allows simultaneous multiparameter analysis of physical and chemical characteristics, evaluating on average more than two thousand particles per second.

What are the characteristics of the equipment?
• Partec brand, CyFlow Space model.
• It has a laser that excites at 488 nm (blue).
• It has three fluorescence emission detectors at 536/40 nm BP (green), 590/50 nm BP (orange) and 630 nm LP (red).
• Software: FloMax.

What are the requirements for samples?
• Minimum quantity required by equipment per sample: 800 µl.
• Concentration: 1 to 5 x 106 cells or particles/ml.
• Size of cells or particles: 1 to 100 µm.
• Cells and/or particles must be broken down.

What are the controls needed to set the equipment?
1. Negative control containing only the supporting medium, without cells and/or particles.
2. Negative control with unmarked cells.
3. Positive controls for each marker separately.
4. Positive controls with all markers together.
